Corrected QT (QTc) interval prolongation is a recognized risk factor for potentially fatal cardiac arrythmias; it is a safety consideration with currently approved menin inhibitors and is listed as a warning in their US Prescribing Information.1–3 Therefore, characterizing the cardiac safety profiles of emerging menin inhibitors is a key aspect of clinical development.1
The investigational menin inhibitor bleximenib is currently being evaluated in the phase I/II cAMeLot-1 (NCT04811560) trial, as monotherapy in patients with relapsed/refractory (R/R) KMT2A-rearranged (KMT2Ar), NPM1-mutated (NPM1m), or NUP98/214-rearranged (NUP98/214r) acute leukemia; and in the phase I/II ALE1002 (NCT05453903) trial, in combination with acute myeloid leukemia (AML)-directed therapies in patients with R/R or newly diagnosed (ND) KMT2Ar, NPM1m, or NUP98/214r AML.
Here, we present a visual abstract outlining analyses of the cardiac safety findings from the cAMeLot-1 and ALE1002 trials, which were presented by Marie Luise Hütter-Krönke at the European Hematology Association (EHA) 2026 Congress.
